Isopentenil pirofosfat (IPP, isopentenil difosfat, atau IDP) adalah senyawa antara dalam jalur reduktase HMG-CoA klasik (umum disebut jalur mevalonat), dan digunakan oleh mikroorganisme dalam biosintesis terpena dan terpenoid. IPP dibentuk dari asetil-CoA melalui jalur mevalonat (bagian "hulu"), dan kemudian diisomerisasi menjadi dimetilalil pirofosfat oleh enzim isopentenil pirofosfat isomerase.

Jalur Mevalonate

Jalur sintesis steroid yang disederhanakan dengan senyawa antara isopentenil pirofosfat (IPP), dimetilalil pirofosfat (DMAPP), geranil pirofosfat (GPP) dan skualena. Beberapa senyawa antara diabaikan. Perlu dicatat skema warna hanyalah sebagai ilustrasi dan tidak secara tepat mewakili asal muasal unit isoprena GPP.

IPP dapat disintesis melalui jalur alternatif yaitu biosintesis isoprenoid tanpa jalur mevalonat, yang terbentuk dari (E)-4-hidroksi-3-metil-but-2-enil pirofosfat (HMB-PP) oleh enzim HMB-PP reduktase (LytB, IspH). Jalur non-mevalonat digunakan oleh banyak bakteri, protozoa apikompleksan seperti parasit malaria, dan plastida tanaman yang lebih tinggi.
