Cromwell terlahir dalam keluarga priayi menengah, tergolong keturunan dari saudari Thomas Cromwell, salah seorang menteri Raja Henry VIII. Hanya sedikit informasi yang diketahui seputar 40 tahun pertama kehidupannya karena hanya empat surat pribadinya yang terlestarikan bersama dengan sebuah ringkasan kata sambutan yang ia sampaikan pada tahun 1628. Ia menjadi seorang Puritan Independen setelah mengalami suatu konversi agama pada tahun 1630-an, menganut suatu pandangan yang relatif toleran terhadap banyak aliran Protestan yang lain pada zamannya. Dikatakan bahwa ia adalah seorang yang sangat religius. Ia terpilih untuk memegang komando dalam kampanye militer Inggris di Irlandia pada tahun 1649–1650. Pasukan Cromwell mengalahkan koalisi Konfederasi dan Royalis di Irlandia serta menduduki negara tersebut, menyebabkan berakhirnya Perang Konfederasi Irlandia.
