Odenkirk lahir di Naperville, Illinois . Dia adalah adik dari aktor Amerika dan komedian Bob Odenkirk, dan bekerja sebagai penulis, produser dan aktor di acara komedi sketsa komedi Mr Show dengan Bob dan David, yang menampilkan saudaranya sebagai lawan mainnya.
